Mikel Leshoure had three touchdowns in the first quarter, helping the Lions get out to a 24-0 lead before the Jaguars first scored.
Matthew Stafford went 22-for-33 for 285 yards.
Blaine Gabbert went 27-for-38 for 220 yards, two touchdowns and two interceptions.
